　　1.用含10%小牛和的RPMI-1640培养液将K562细胞配成1&time105/ml浓度。用同样培养液将MC配成1&time106/ml浓度。

　　2.取96孔圆底，每孔加0.1ml K562细胞悬液;每孔加适量MC使效靶比为1：1～5：1，每种处理3个复孔;3个对照孔只加效应细胞;8个对照孔只加靶细胞;4个对照孔只加细胞培养液。每孔加20μl Alamar Blue;每孔总量为0.2ml。

　　3.在37℃ 5% CO2的饱和水汽二氧化碳培养箱中培养24小时。

　　4.直接在荧光检测仪上检测荧光强度，激发波长530nm，发射波长590nm。各孔荧光强度值应减去培养液对照荧光强度的平均值，各复孔荧光强度的平均值记为AF。按下式计算特异性杀伤百分比：
